Barnet boss Paul Fairclough made three changes to his starting line-up following Tuesday night's defeat against SwindonTown.
Tresor Kandol replaced Magno Vieira in the Bees attack, whilst Nicky Nicolau made his debut at left back in place of Adam Gross. There was also a debut on the right side of midfield for Barry Cogan, who signed from Millwall on Thursday - Richard Graham making way with Nicky Bailey switching over to the left of the Bees midfield and Sinclair to the centre to accommodate Cogan in the side.
Barnet started the game the brighter of the two sides and nearly took the lead in bizarre fashion after just two minutes when a long looping ball forward from his own half by Ian Hendon saw the ball bounce in the six yard box and hit the top of the crossbar.
Just a minute later and the Bees came close again when a low cross into the box by Bailey was fumbled on the line by Stanley goalkeeper Ian Dunbavin but Sinclair was unable to force the ball home and Michael Welch cleared to safety.
Barnet continued to play the better football in the opening stages of the game and they again came close in the 13th minute to breaking the deadlock when a Cogan corner bounced around in the area but Sinclair could only stab his effort wide.
A minute later and the same two players combined once more but, although this time hitting the target, Sinclair's effort lacked power and Dunbavin easily held onto the ball.
Despite the Bees dominance though, it was the home side who could have taken an undeserved lead in the 16th minute when a deep cross by Leam Richardson found Welch unmarked at the back post but the big centre back, still up front from an earlier free kick, could only direct his header wide of Lee Harrison's right hand post.
Two minutes later and Stanley again wasted a good chance when Paul Mullin could only direct his header from six yards straight at Harrison following a fine Gary Roberts cross from the right.
Ian Craney then shot wide in the 23rd minute after fine play by Romauld Boco although it was Barnet who were having the greater possession of the ball.
And that greater possession nearly told when a fine ball forward by Andy Hessenthaler after 26 minutes found Kandol in the box but the strikers shot from the left lacked power and Dunbavin gathered comfortably.
Williams then headed high over the crossbar at the other end for Accrington but they then went closer when Roberts hit the post after 32 minutes from four yards after Paul Mullin squared the ball from the Stanley right.
Kandol then curled an effort wide for the Bees as they continued to look dangerous going forward but Stanley went straight down the other end to take the lead in the 35th minute when Boco raced onto a long ball forward, Warhurst misjudged his header, and the Stanley front man calmly lobbed the ball over Harrison from 10 yards to send the home side into the lead.
Barnet almost equalised immediately when a Kandol flick on found Hatch but Dunbavin just managed to claim the ball ahead of the Bees striker.
In a reversal of the first half, it was Accrington who began the second period the brighter of the two sides and just two minutes in, Craney's 25 yard strike was pushed over the crossbar by Harrison for a corner kick.
Two minutes later and again Stanley threatened when the scorer of the first goal, Boco shot over from 25 yards after the Bees failed to clear.
Straight down the other end and Kandol had a great chance to equalise but he missed the ball completely after Bailey beat his man down the left and crossed low into the penalty area.
Again the Bees were made to pay for that miss as Accrington went straight down the other end to get their second goal of the game and again it was that man Boco.
Roberts kept the ball in on the Bees left, forcing Harrison into a great save but Boco was there to convert the rebound from six yards to send the home fans into delirium and the travelling Bees into a state of shock.
Bailey shot just wide for Barnet from an acute angle on the left after 62 minutes as they looked to reduce the deficit as Stanley looked to retain their two goal lead.
Fairclough then made a double substitution in the 72nd minute - Magno Vieira replacing Liam Hatch in the Bees attack and Anthony Charles taking the place of Captain Ian Hendon at right back.
The changes though seemed to inspire Accrington rather than Barnet and the home side came close to scoring twice in the matter of seconds.
Firstly, Boco advanced towards goal after a long pass forward but after outpacing Nicolau the striker could not claim his hat-trick as he directed his effort over the crossbar. Then, after a Harrison goal kick, Stanley won the ball back, and Craney's cross from the right swerved in the wind forcing Harrison to tip the ball over his crossbar for a corner kick.
Stanley were now playing some fine one touch football and Andrew Todd had a chance in the 78th minute to get the home sides third but his effort from the right of the penalty area lacked power and was held by Harrison.
Ten minutes from time and Fairclough made his third and final substitution with debutant Cogan being replaced by Graham in the Bees midfield.
In truth though, the Bees were looking like a beaten side at this stage, and despite the effort, they were struggling to force Stanley goalkeeper Dunbavin into action.
Two minutes from time though and the Bees did get a goal back when Bailey worked his way into the box before forcing Dunbavin into a fine save but Simon King was there to blast the rebound home from eight yards to reduce the deficit.
Nicolau then saw his low shot held by Dunbavin a minute later as the Bees looked to steal a late equaliser.
The referee added three minute of added time and a minute in, Graham shot inches wide from 12 yards after Kandol flicked on a long Harrison free kick.
However, there was not enough time left for Barnet to find that crucial second goal as Accrington Stanley won their first football league game for 44 years.
